With the Sequencher® software, for sequence variant analyses, we used the Seg-1 sequence subtype of GenBank accession number [19507058](https://www.ncbi.nlm.nih.gov/nuccore/19507058), NCBI Reference Sequence: [19507058](https://www.ncbi.nlm.nih.gov/nuccore/19507058), which is identical to the sequence of *Ulva australis* (L.) Ach. ex J. Agardh, 1801.0 (GenBank accession no. [HM744110.1](https://www.ncbi.nlm.nih.gov/nuccore/HM744110.1)).
We conducted neighbor joining (NJ) analysis using MEGA5.05 software () to determine the evolutionary relationships among three ITS variants and three ETS variants. The Kimura 2-parameter model in MEGA5.05 was chosen to perform the NJ analysis. Bootstrap (BS) analysis was performed to estimate the reliability of the branching analysis, which was conducted with 1000 replicates.
The structure and optical rotation of the compound were established by spectral analyses (^1^HNMR, ^13^CNMR and MS spectra) of the compound isolated from the whole plant of Ulva sp. 2, and direct evidence was obtained by preparing the crystals of reticuloside B by recrystallization from an absolute ethanol/water solution.
